ABSTRACT
Nowadays , determination of hazardous and toxic pollutants is one of the most critical challenges whole over the world. Heavy metals attracts a lot of attention among other pollutants and lead is and one of the most dangerous heavy metals . than determination of it needs a great deal of financial supports and of course , a lot of time . Although there has been successful efforts to determine lead by atomic absorption spectroscopy , but in this study we tried hard to set up a procedure which helps us to determine lead by using routine AAS instruments , which exists in most of analytical laboratories.
INTRODUCTION
Lead is a toxic metal whose widespread use has caused extensive environmental contamination and health problems in many parts of the world . Human exposure to lead is estimated to account for 143 000 deaths every year and 0.6% of the global burden of disease (1). Lead is a cumulative toxicant that affects multiple body systems, including the neurological, haematological, gastrointestinal, cardiovascular and renal systems.(2) Today, laboratories primarily assess lead exposure with whole blood lead measurements. Although a number of other human tissues and fluids, such as hair, teeth, bone and urine, also reflect lead exposure, the concentration of lead in whole blood has gained wide acceptance as the most useful tool for screening and diagnostic testing(3,4). Although there are some other alters to determine lead concentration in blood like Flame atomic absorption spectrophotometry (FAAS) or Anodic stripping voltammetry (ASV) , optical emission , polarographic , spectrophotometric and Atomic fluorescence( table 1)(5-23) , but one of the most common laboratory method to determine blood lead concentration is graphite furnace atomic absorption spectrophotometry(GFAAS) and this choice was greatly due to its good sensitivity .
چکیده
امروزه تعیین آلودگی های خطرناک و سمی یکی از مهم ترین چالش های موجود در سراسر جهان است. فلزات سنگین توجه زیادی را در میان سایر آلاینده ها جذب می کنند و سرب یکی از فلزات سنگین ترین فلزات است. از تعیین آن نیاز به مقدار زیادی از حمایت مالی و البته، بسیاری از زمان. اگرچه تلاش های موفق برای تعیین سرب با استفاده از طیف سنجی جذب اتمی صورت گرفته اما در این مطالعه تلاش شده است تا روش هایی را ایجاد کنیم که به ما کمک می کند تا با استفاده از ابزارهای معمول AAS، که در اکثر آزمایشگاه های تحلیلی وجود دارد، تعیین شود.
مقدمه
سرب یک فلز سمی است که استفاده گسترده آن باعث آلودگی محیط زیست و مشکلات بهداشتی در بسیاری از نقاط جهان شده است. قرار گرفتن در معرض سرب انسان به علت 143000 مرگ و میر در سال و 0.6٪ از بار جهانی بیماری (1) تخمین زده می شود. سرب یک سموم تجمعی است که بر سیستم های متعدد بدن، از جمله سیستم های عصبی، هماتولوژیکی، دستگاه گوارش، قلب و عروق و کلیه تاثیر می گذارد. (2) امروزه آزمایشگاه ها به طور عمده ارزیابی سرب را با اندازه گیری کل سرم خون ارزیابی می کنند. گرچه تعدادی از دیگر بافت ها و مایعات های انسانی مانند مو، دندان، استخوان و ادرار نیز منعکس کننده سرب هستند، غلظت سرب در کل خون، به عنوان ابزار مفید برای غربالگری و آزمایش های تشخیصی (3،4 ) اگر چه تغییرات دیگری در تعیین غلظت سرمی خون مانند اسپکتروفتومتری جذب اتمی فلامن (FAAS) یا ولتامتریت سدسازی آنودایز (ASV)، انتشار نوری، فلورسانس طیفی، اسپکتروفتومتری و فلورسانس اتمی وجود دارد (جدول 1) (23-23)، اما یکی از شایعترین روش آزمایشگاهی برای تعیین غلظت سرب خون، اسپکتروفتومتری جذب اتمی کوره گرافیتی (GFAAS) است و این انتخاب به دلیل حساسیت خوب آن است.
