چکیده
هدف اصلی از انجام این مطالعه تهیه نانو ذرات کیتوسان بارگذاری شده با دارو و بررسی اثر این ذرات بر خواص فیزیکی- مکانیکی و خواص میکروبی هیدروژلها بر پایه پلی وینیل الکل بود . بدین منظور نانو ذرات کیتوسان با روش ژل شدن یونی تهیه شدند. نتایج حاصل از مشاهدات میکروسکوپ الکترونی روبشی، تهیه موفق نانو ذرات کیتوسان کروی با متوسط اندازه ذرات 65 nmرا تایید کردند. هیدروژل PVAحاوی داروی خالص تتراساکلین و نانو ذرات کیتوسان بارگذاری شده با این دارو، با روش پرتودهی الکترونی تحت دز 25تهیه شدند. نتایج حاصل از آزمون تورم و بررسی های سینتیک تورم حاکی از کاهش قابل توجه میزان نفوذ پذیری و در نتیجه پدیده تورم ناشی از حضور نانو ذرات در زمینه هیدروژل بودند، اگر چه خواص مکانیکی آن چنان تحت تاثیر نانو ذرات قرار نگرفتند. خواص میکروبی از هیدروژل حاوی داروی مستقیم یا نانو ذرات بارگذاری شده با دارو نشان دهنده بهبود خواص آنتی باکتریال آنها بود، البته هیدروژلها حاوی نانو ذرات کیتوسان خواص ضد میکروبی به مراتب نشان دادند.
مقدمه
در سالهای اخیر، پیشرفتهای قابل توجهی در استفاده از پلیمرهای زیست پزشکی قابل تورم در آب به عنوان زخم پوشهای حامل دارو انجام گرفته است. در واقع هیدروژلها، به دلیل زیست سازگاری بالا، محتوای زیاد آب، کشش سطحی کم در سطح مشترک با پوست و خواص مکانیکی مشابه این ژلها با بدن، مورد استفاده به عنوان زخم پوش قرار گرفته اند. منافذ موجود در هیدروژلها باعث می شود تا بتوان دارو را به راحتی درون آنها قرار داد و به عنوان یک سامانه دارورسانی به کار برد. اما بارگذاری مستقیم دارو در زمینهی هیدروژل، غالباً منجر به سرعت نسبتاً بالای رهایش دارو از آن میشود، همچنین تخریبهای محیطی برای دارو را نیز محتمل میسازد. استفاده از نانوذرات برای بارگذاری دارو، ضمن کمک به کنترل سنتیک رهایش دارو از هیدروژل، به عنوان محافظی برای دارو در برابر عوامل محیطی محسوب میشود.
ABSTRACT
The main objective of this study was to prepare nanosized chitosan particles loaded with drugs and to investigate the effect of these particles on physical-mechanical properties and microbial properties of hydrogels based on polyvinyl alcohol. For this purpose, chitosan nanoparticles were prepared by ion gelling method. The results of scanning electron microscopy observations confirmed the successful preparation of spherical quothuclino nanoparticles with an average particle size of 65 nm. PVA hydrogel containing a pure tetracycline drug and chitosan nanoparticles loaded with this drug were prepared by electron beam irradiation under a dose of 25. The results of the swelling test and the kinetic studies of swelling indicated a significant reduction in permeability and as a result of the inflation phenomenon caused by the presence of nanoparticles in the field of hydrogel, although its mechanical properties were not so affected by the nanoparticles. The microbial properties of the direct drug containing hydrogel or the nanoparticles loaded with the drug showed an improvement in their antibacterial properties, although hydrogels containing chitosan nanoparticles exhibited antimicrobial properties.
INTRODUCTION
In recent years, significant advances have been made in the use of swollen biomedicine polymers in water as drug carrier wounds. In fact, hydrogels have been used as wounds due to high biocompatibility, high water content, low surface tension on the skin surface, and mechanical properties similar to those of the body gels. The pores in the hydrogels make it easy to put the drug into them and use it as a drug delivery system. However, direct loading of the drug in the field of hydrogel often leads to a relatively high rate of drug release, and also makes possible environmental degradation for the drug. The use of nanoparticles for drug loading, while helping to control the release of the drug, is hydrogel, as a protective medicine against environmental factors.
