ABSTRACT
In multicellular organisms the fusion of two gametes with a haploid set of chromosomes leads to the formation of the zygote, the frst cell of the embryo. Accurate execution of the meiotic cell division to generate a female and a male gamete is required for the generation of healthy offspring harboring the correct number of chromosomes. Unfortunately, meiosis is error prone. This has severe consequences for fertility and under certain circumstances, health of the offspring. In humans, female meiosis is extremely error prone. In this chapter we will compare male and female meiosis in humans to illustrate why and at which frequency errors occur, and describe how this affects pregnancy outcome and health of the individual. We will frst introduce key notions of cell division in meiosis and how they differ from mitosis, followed by a detailed description of the events that are prone to errors during the meiotic divisions.
INTRODUCTION
Diploid organisms contain two copies of the genome, originating from the two parents and organized as thread-like structures named chromosomes. In S-phase, which precedes cell division in somatic and germ cells, paternal and maternal genomes are replicated, the cell harbors therefore four copies of the genome. As cells enter mitosis, DNA condensation takes place, and chromatids, each corresponding to one copy of the genome, become visible. Two sister chromatids of the same parental origin are paired and segregated to generate two identical daughter cells. In meiosis on the other hand, four cells each harboring only one copy of the genome are produced to form the haploid gametes (Fig. 1.1). In female meiosis, only one of these four cells develops into an oocyte, the remaining cells degenerate as Polar Bodies (PBs). The generation of haploid gametes is possible due to the specialized segregation pattern of the meiotic cell division which ensures that each gamete receives one chromatid of each chromosome pair . Contrary to mitosis, the aim of meiosis is not to generate identical daughter cells, but to recombine the maternal and paternal genome to generate new genetic variations in the offspring. This requires the pairing of homologous chromosomes from different parental origin, and DNA recombination, which take place in meiotic prophase, before the meiotic divisions. When germ cells enter the meiotic cell division, recombination events are fnalized.
چکیده
در ارگان های مولکولی همجوشی دو گامت با مجموعه ای از هپلوئید کروموزوم منجر به تشکیل زگوت، سلول پرنده جنین می شود. اعدام دقیق از تقسیم سلول های مایوتیک برای تولید یک زن و یک مرد نر و ماده برای تولید فرزندان سالم و داشتن تعداد مناسب کروموزوم لازم است. متاسفانه، مايوز خطا مستعد است. این عواقب شدید برای باروری و تحت شرایط خاص، سلامت فرزندان است. در انسان، مايع زن ماده بسيار خطرناک است. در این فصل مائویسم مرد و زن را در انسان مقایسه خواهیم کرد تا نشان دهد که چرا و چه خطایی در فرکانس اتفاق می افتد و توضیح دهید که چگونه این پیامد حاملگی و سلامت فرد را تحت تاثیر قرار می دهد. ما از مفاهیم کلیدی تقسیم سلولی در میزیوز ارائه خواهیم کرد و این که چگونه آنها با میتوز متفاوت است، به دنبال توضیح مفصلی از حوادثی که مستقیما به خطاها در تقسیم های مایع می پردازند.
مقدمه
ارگانهای Diploid حاوی دو نسخه از ژنوم است که از دو پدر و مادر تشکیل شده و به عنوان ساختارهای مشابه به نام کروموزوم سازماندهی شده اند. در مرحله S که قبل از تقسیم سلولی سلول های جسم و جسمی، ژنوم های پدر و مادر تکرار می شوند، سلول به این ترتیب چهار نسخه ژنوم را در اختیار دارد. همانطور که سلول ها به میتوز وارد می شوند، تراکم DNA اتفاق می افتد، و کروماتید ها، هر کدام به یک کپی از ژنوم، قابل مشاهده می شوند. دو کروماتید خواهر از همان منشاء والدین برای تولید دو سلول دخترانه یکسان زوج و جدا می شوند. از سوی دیگر، در م یوزیس، چهار سلول که تنها یک نسخه از ژنوم را پوشش می دهند تولید می شوند تا گامت های هپلوئید ایجاد شود (شکل 1.1). در مايوز زن، فقط يكي از اين چهار سلول به تخمك تبديل مي شود، سلول هاي باقيمانده به عنوان بدن هاي قطبي (PBs) دژنراسيون مي كنند. تولید ژنهای هپلوئید ممکن است به علت الگوی جداسازی تخصصی بخش تقسیم سلولی مئوتیک باشد که تضمین می کند که هر یک از گلوتامین یک کروماتید از هر جفت کروموزومی دریافت می کند. بر خلاف ميتوز، هدف مايوز توليد سلول هاي يكسان نيست، بلكه مجددا ژنوم مادر و پدري را براي ايجاد تغييرات ژنتيكي جديد در فرزندان بازسازي مي كند. این امر نیاز به جفت شدن کروموزومهای همولوگ از مشتقات والدین مختلف و نوترکیب DNA دارد، که در پروپаاز مايوکتیو قبل از تقسیم بندی های مايوتيک انجام می شود. هنگامی که سلول های بنیادی به بخش سلول های مايوکتیو وارد می شوند، حوادث نوترکیبی فنانالیزه می شوند.
