ABSTRACT

Stem cells are self-renewing and pluripotent, meaning that they are capable of continuous proliferation and  terminal differentiation into various cell types. Stem cells are classifed as either embryonic or adult based on their origin, and give rise to various organs and tissues (Thomson et al. 1998; Shamblott et al. 1998). Recent studies indicated that induced pluripotent stem cells (iPSCs) and directly induced neurons are included in the category of stem cells (Takahashi and Yamanaka 2006; Pang et al. 2011). Stem cell transplantation has enabled powerful new therapeutic strategies in research for the treatment of various human neurological diseases such as Alzheimer’s disease (AD) and Huntington’s disease (HD) (Kim et al. 2013; Nikolic et al. 2008). The identifcation, generation, and optimization of suitable stem cell types for cell therapy is necessary for the full utilization of this promising therapeutic approach in neurological disease (Kim et al. 2013). In this chapter, we review the utility and limitations of different stem cell types and discuss recent advances in the therapeutic use of stem cells in neurological and neurodegenerative disease.

INTRODUCTION

  Stem cells are undifferentiated cells that are capable of symmetrical and/or asymmetrical division by mitosis, and are also able to ifferentiate into mature tissuespecifc cells (Gage and Temple 2013; Martello and Smith 2014; Golas and Sander 2016). Stem cells have been used for the treatment of various neurological diseases including AD and HD; cells for these purposes have been derived from embryonic stem cells (ESCs), induced pluripotent stem cells, and tissue-specifc adult stem cells (Hargus et al. 2014; Maucksch et al. 2013; Schwarz and Schwarz 2010).  Human ESCs (hESCs) are pluripotent cells derived from the inner cell mass (ICM) of the blastocyst. hESCs can differentiate into all cell types from three embryonic germ layers: the endoderm, mesoderm and ectoderm (Thomson et al. 1998). Accordingly, these cells can be applied therapeutically in human disease states (Nishio et al. 2016). Although hESCs initially require culture on a feeder layer of mouse embryonic fbroblasts and animal serum (Thomson et al. 1998), many studies have established animal substance-free, feeder-free hESC culture conditions for clinical applications (Hovatta et al. 2014). Because hESCs are derived from in vitro fertilized blastocysts donated for research, the transplantation of hESCs faces ethical and legal limitations (Hovatta et al. 2010; Lo et al. 2005).

چکیده

سلول های بنیادی خود تجدید می شوند و قادر به تکامل هستند، به این معنی که آنها قادر به تکثیر و تمایز پایدار به انواع سلول های مختلف هستند. سلول های بنیادی به صورت جنینی یا بالغ بر مبنای مبدأ آنها طبقه بندی می شوند و باعث ایجاد اندام و بافت های مختلف می شوند (Thomson et al. 1998؛ Shamblott et al.، 1998). مطالعات اخیر نشان می دهد که سلول های بنیادی پلوروپتوپ القایی (iPSCs) و نورون هایی که به طور مستقیم القا شده اند، در رده سلول های بنیادی قرار دارند (Takahashi و Yamanaka 2006؛ Pang et al.، 2011). پیوند سلول های بنیادی روش های جدید درمان دارویی جدید را در درمان بیماری های مختلف بیماری های عصبی مانند بیماری آلزایمر (AD) و بیماری هانتینگتون (HD) (Kim et al. 2013؛ Nikolic et al. 2008) فراهم کرده است. شناسایی، تولید و بهینه سازی انواع سلول های بنیادی مناسب برای درمان سلول ها برای استفاده کامل از این روش درمانی نورپرداز ضروری در بیماری های عصبی ضروری است (Kim et al.، 2013). در این فصل، ما ابزار و محدودیت های انواع مختلف سلول های بنیادی را مورد بررسی قرار می دهیم و پیشرفت های اخیر در استفاده از سلول های بنیادی در بیماری های نورولوژیکی و نوروژنرژیک را مورد بحث قرار می دهیم.

مقدمه

سلول های بنیادی سلول های نامتقارن هستند که قادر به تقسیم متقارن و / یا تقسیم نامتقارن توسط میتوز هستند و همچنین می توانند سلول های بافتی بالغ را جذب کنند (Gage and Temple 2013؛ Martello and Smith 2014؛ Golas and Sander 2016). سلول های بنیادی برای درمان انواع بیماری های عصبی مانند AD و HD استفاده شده است. سلول ها برای این اهداف از سلول های بنیادی جنینی (ESCs)، سلول های بنیادی پلوروپتوژن القا شده و سلول های بنیادی بالغ بافت مشخص شده است (Hargus et al. 2014؛ Maucksch et al. 2013؛ Schwarz and Schwarz 2010). ESC های انسانی (hESCs) سلول های انعقادی هستند که از توده سلولی داخلی (ICM) بلستوسیست منتقل می شوند. hESCs می تواند به تمام انواع سلول از سه لایه ژرمین جنینی تمایز یابد: اندودرم، مزودرمان و اکوترم (Thomson و همکاران، 1998). بر این اساس، این سلول ها می توانند در شرایط بیماری های انسانی درمان شوند (Nishio et al.، 2016). اگر چه hESC ها ابتدا نیاز به کشت در لایه فیدر فیبروبلاست های جنینی موش و سرم حیوانات دارند (Thomson et al.، 1998)، مطالعات بسیاری در زمینه شرایط کشت بدون hid، بدون فیدر حیوانات برای برنامه های بالینی ایجاد کرده اند (Hovatta et al.، 2014). از آنجا که hESC ها از بلاستوسيست هاي بذر در in vitro به منظور تحقيق اهدا شده اند، پيوند hESC با محدوديت هاي اخلاقي و قانوني مواجه است (Hovatta et al. 2010؛ Lo et al 2005).
