ABSTRACT
Scientific advances have significantly improved the practice of medicine by providing objective and quantitative means for exploring the human body and disease states. These innovative technologies have already profoundly improved disease detection, imaging, treatment and patient follow-up. Today’s analytical limits are at the nanoscale level (one-billionth of a meter) enabling a detailed exploration at the level of DNA, RNA, proteins and metabolites which are in fact nano-objects. This translational review aims at integrating some recent advances from micro- and nano-technologies with high potential for improving daily oncology practice.
INTRODUCTION
Nanotechnology (from nano, ‘dwarf’ in Greek) was envisioned by the physicist and Nobel laureate Richard Feynman in his seminal lecture “there is plenty of room at the bottom” in 1959 . It provides scientists with new tools for the investigation of objects in the range of -typically but not exclusively- 1 nm to 100 nm: the typical scale of biologically relevant molecules like proteins, enzymes, receptors, haemoglobin and antibodies for instance (see Fig. (1)). Nanotechnology, coined in 1974, refers to the handling and/or engineering of nano-objects on the scale of molecules , and brings at least 2 major attractive features to existing techniques. Synthetic nanoscale materials are inherently small, with at least one dimension in the 1- 100nm range so they can cross biological barriers including the blood-brain barrier , transit out blood vessel walls or the cell membrane by different uptake mechanisms . They can therefore interact with a wide panel of biological entities (e.g.: from proteins to cells). Also, nano-objects exhibit tuneable physical (e.g.: electrical, magnetic, optical ,mechanical), chemical (e.g.: reactivity , melting point or biological properties that are dramatically different from the same materials in larger scale forms due to modified quantum mechanical properties. Depending on the type of applications, nano-objects can span different shapes (e.g.: nanospheres, nanotubes, nanorods, nanowires) and materials (e.g.: silica, gold, semiconductors, iron oxides, lipid- or polymer-based) so they can be hollow, porous or solid. These characteristics confer nanoobjects high interaction and transport capabilities, making them attractive tools for the design of biosensors, imaging contrast agents and/or therapeuti ccarriers.
چکیده
پیشرفت های علمی به طور قابل توجهی بهبود عمل طب را با ارائه ابزاری عینی و کمی برای کشف بیماری های بدن انسان و بیماری ها. این فن آوری های نوآورانه در حال حاضر عمیقا بهبود بیماری، تصویربرداری، درمان و پیگیری بیمار را بهبود بخشیده است. محدودیتهای تحلیلی امروز در سطح نانومقیاس (یک میلیاردمتری از یک متر) امکان بررسی دقیق در سطح DNA، RNA، پروتئینها و متابولیتها هستند که در واقع نانو اشیا هستند. این بررسی انتقادی با هدف ادغام برخی پیشرفت های اخیر از میکرو و فناوری نانو با پتانسیل بالایی برای بهبود عمل روزانه انکولوژی انجام شده است.
مقدمه
فناوری نانو (از نانو، ‘کوتوله’ در یونانی) توسط فیزیکدان و برنده جایزه نوبل ریچارد Feynman در سخنرانی معاصر او “اتاق زیادی در پایین” در سال 1959 پیش بینی شده بود. این دانشمندان ابزار جدیدی را برای بررسی اشیا در محدوده های معمول، اما نه به طور انحصاری- 1 نانومتر تا 100 نانومتر فراهم می کنند: مقیاس نمونه ای از مولکول های مرتبط با زیست شناسی مانند پروتئین، آنزیم ها، گیرنده ها، هموگلوبین و آنتیبادی ها به عنوان مثال (نگاه کنید به شکل.
